Anthony Cooper ist John Locke's biologischer Vater. who, when found by John, was on dialysis and in need of a new kidney. He got close to John by taking him hunting occasionally, and eventually conned him out of a kidney in a ploy using Locke's mother Emily.

Theories
- Anthony Cooper may actually be the notorious D.B. Cooper. He's the right age, a meticulous planner and certainly ruthless enough.
- Anthony Cooper could be Radzinsky. This explains two parts of the mystery. Cooper stole money from the mafia members that approached Locke after his father faked his death. He then kept some by filtering it through DHARMA, in turn they hid him on the island in the Swan. The Sickness could have caused his kidney ailment and so the DHARMA people faked his sucide when Kalvin was away from the hatch and gave a fake suicide note (Cooper is willing to do anything to get what he wants) and then took him back to the states where he recieved a large sum of money. This is why Locke is so willing to stop pushing the button because he finds other evidence that his father was in the hatch and Locke refuses to become his father, but at first he was willing to do anything for his love (thus he pushes the button vigilantly).
- Anthony Cooper could in fact be the real Mr. Sawyer.
- On the Season 2 DVD set, under the "Lost Connections" feature, there's a curious link between Locke and Sawyer that plays a clip from Season One where Locke asks Sawyer why he chose that name. This seems to suggest that Locke's father is indeed the real Sawyer.
- Could be a Dharma Recruiter. Seemed to initiate events that lead to Locke ending up on the island, and if the theory that he's the real Sawyer is true, then Sawyer as well.
- Is Him. (When Helen meets him for the first time, she asks "Are you him?")

- Is not Lockes father at all, as Emily Annabeth Locke's claims of John being immaculately conceived could be the truth. The lack of blood links between the two, anyway, wouldn't explain why Cooper was looking specifically for Locke's kidney.
- Perhaps Locke's private investigator had been compromised, and Anthony led Locke to believe he was his father so he could take his kidney.
- Anthony may have married and had other children he didn't raise besides Locke.
